Simultaneous Separation and Determination of Four Fluoroquinolones by HPLC Employing a New Phenylala

  In the present work, a new phenylalanine-substituted calixarene bonded silica stationary phase (PHE-Cx4) was synthesized, structurally characterized, and employed to separate four fuoroquinolones.A simple, rapid and sensitive liquid chromatographic method was developed and validated for simultaneous separation and determination of four fluoroquinolones (lomefloxacin hydrochloride, ofloxacin, enrofloxacin, sarafloxacin hydrochloride).The optimum experimental conditions have been studied carefully, and good linear range over the concentration range of 20-500 ng/mL for Lomefloxacin hydrochloride, 25-700 ng/mL for Ofloxacin, 30-800 ng/mL for enrofloxacin and 30-1000 ng/mLfor sarafloxacin hydrochloride were obtained.The limits of detection (LOD) were found to be 7.0 ng/mL, 8.5 ng/mL, 9.2 ng/mL and 9.9 ng/mL, for lomefloxacin hydrochloride, ofloxacin, enrofloxacin and sarafloxacin hydrochloride, respectively.Intra-day and inter-day relative standard deviation (RSD) at three different concentrations were determined.The low RSD values and high recovery values indicate good precision and accuracy of the proposed methods.The column has been successfully employed for the simultaneous analysis of the compounds extracted from milk, eggs, meat and water.
