A kind of "two-dimensional stationary phase (2DSP)" of liquid chromatography was first synthesized, which has the characters of both weak cation-exchange chromatography and hydrophobic interaction chromatography.The 2DSP is composed of four types of groups including three secondary amines, one thiol, two carboxyls, and silane group.The excellent performance of the synthesized stationary phase is attributed to the "groups-groups" dynamic matching, that is, "apolar-apolar", "polar-polar", and "charge-charge" interactions that exist on the contact region between the stationary phase and protein molecules.Only by controlling the salt concentration of the mobile phase of hydrophobic chromatography, five and eight standard proteins can be completely separated under IEC or HIC mode with the single column (2DLC-1C), respectively.It was found that the dynamic adsorption capacity of the stationary phase for lysozyme was 17.27mg/g and 23.79mg/g under IEC and HIC modes, respectively.Furthermore, the mass recovery of several test proteins under the two modes was found to be higher than 96%.The foregoing result indicates that in comparison with conventional weak cation-exchange and hydrophobic interaction chromatography stationary phases, the synthesized 2DSP has a superior performance.
