抗癌止痛肽镇痛作用的钠离子通道机制

  目的 探讨抗癌止痛肽(AGAP)的止痛作用机制,考察了其镇痛作用是否与调节钠离子通道相关,以期为阐述抗癌止痛肽的镇痛作用与机制提供理论依据。方法 应用原代培养和急性分离的方法获得大鼠背根神经节(DRG)神经元,应用MTT法考察AGAP对大鼠DRG神经元存活率的影响;采用膜片钳全细胞记录模式结合分子生物学实验考察抗癌止痛肽的镇痛作用的离子通道机制。结果 实验结果表明,抗癌止痛肽对原代培养和急性分离的大鼠DRG神经元钠离子通道电流均能显著抑制。进一步分型研究结果表明,抗癌止痛肽能以浓度依赖和电压依赖的方式显著抑制TTX-R Na+电流,使TTX-R Na+的激活曲线向负向移动。进一步的研究结果表明,抗癌止痛肽对急性分离的CCI模型大鼠DRG神经元Na+电流具有显著地抑制作用。结论 提示抗癌止痛肽的镇痛作用可能是通过调节DRG神经元钠离子通道的实现的。
