BD now has a family of 4 violet-excitable fluorophores, BD Horizon V450, V500, V535, and AmCyan, comprising both organic dyes and fluorescent proteins.All of these fluorophores allow the synthesis of functional antibody conjugates.These antibody conjugates are compatible with multi-laser flow cytometry, requiring only the cytometer employed have a violet laser with an emission line at or near 405 nm.The fluorophores have their excitation maxima in the range from 400 to 458 nm, and their emission maxima from 450 to 535 nm.These 4 fluorophores can be used for polychromatic flow cytometry in 3 different 2-color combinations using a single configuration of dichroic mirrors and bandpass filters, regardless of which combination is chosen.Two of the fluorophores, V500 and AmCyan, fill a spectral opening which has yet to be taken advantage of by other manufacturers of flow cytometry reagents.A reengineered AmCyan has eliminated virtually all blue-laser excitation, which previously led to excessive spillover into the fluorescein-channel.
