目的:观察miR-21对缺氧复氧(H/R)诱导H9C2细胞凋亡的保护作用及探讨其可能机制.方法:将细胞随机分为:①vehicle control;②抑制物阴性对照(inhibitors NC);③模拟物阴性对照(mimics NC);④H/R+vehicle control;⑤H/R+inhibitors NC;⑥H/R+mimics NC;⑦H/R+miR-21抑制剂(inhibitors);⑧H/R+miR-21模拟物(mimics).H/R处理将细胞低氧培养4小时后常氧培养3小时.将50nM的miR-21inhibitors及50nM的miR-21mimics用LipofectamineTM2000转染至H9C2细胞建立表达抑制及过表达miR-21细胞模型.PTEN是miR-21靶基因,以PTEN/AKT途径作为拟观察的miR-21下游通路;Bax/Bcl-2、caspase-3为凋亡指标.Realtime PCR检测miR 21水平,Western印迹方法检测Bax/Bcl-2、caspase-3、PTEN、p Akt的蛋白水平,流式细胞仪检测细胞凋亡率.
