  CdGAP (Cdc42 GTPase-activating protein) is a negative regulator of the small GTPases Racl and Cdc42,members of the Rho family of small GTPases.These proteins play a crucial role in many cytoskeletal-dependent cellular processes such as cell migration and adhesion,morphogenesis,cell cycle progression,gene expression,and apoptosis.Each of these cellular functions plays an active role in the development and progression of cancer.Rho GTPases act as molecular switches and exist in either an inactive GDP-bound or an active GTP-bound conformation.This GDP/GTP cycle is regulated by at least three families of proteins such as Guanine nucleotide exchange factors (GEFs),GTPase-activating proteins (GAPs),and guanine nucleotide dissociation inhibitors (GDIs).In cancer cells,the cytoskeletal architecture is altered by the actions of oncoproteins,or loss-of-function of tumor suppressor proteins.As Rho proteins are major determinants of cytoskeletal remodeling,their regulators and effectors are both crucial for normal cellular responses.Here I will discuss the molecular mechanisms underlying the regulation and cellular functions of CdGAP,a potential target for therapeutic agents against cancer.
