The establishment of tree shrew glioma model and analysis of pathological feature

  Glioblastoma is one of the most harmful and fateful human brain cancer with aggressive disease progress and slim chance of surviving more than 3 year.So,either new drug test or new therapy strategy is hardly needed and both of them are relied on animal model.We selected tree shrew,the promising animal model,to attempt to establish an ideal model and mimicthe pathological feature of glioma in human.Firstly,we construct the lentivirus vector which overexpress Ras gene and knockdown p53gene at the same time.The strategy of vector construction is based on the data from Genomic studies of glioblastoma.In addition,we detect the expression effiency of target genes to make sure the vector is efficient.Then the vector experienced the lentivirus package with the help of packaging plasmidsand virus titer determination.Aided by the brain stereotaxic apparatus,virus was precise injected into the hippocampus of tree shrew.All of the tree shrews appeared glioma symptoms and died finally,the most sensitive tree shrew died in 39 days after injection.Dissection showed the apparent pathogenic change in hippocampus,further paraffin section and HE staining revealed the typical character of glioma,including cell necrosis,invasion,rapidly proliferation,blood vessel hyperplasia,which are similar to those in human.
