1.The Research Follow on tyrosine kinase receptor inhibitor in clinic base has been conducted.We constructed both first targeting to tolic aid receptor method and ZD6474 with minicircle DNA carrying thereuputic gene as nano cell therapy for second target of cancer.It can be specific target to the tolic aid highly expressed cancer cell,and xenografted model as well.The anti-tumor effect of ZD6474-minicircle-IFN-r was a significantly higher than minicircle IFN-r/ZD6474 alone,the later has been in phase Ⅲ clinical trial in Europe and USA.Further investigation on ZD6474 inhibiting SrC Kinase leading to apoptosis of imatinib-resistaut K562 cells.It demonstrated that overactivited anti-tumor effects of ZD6474 were due to its inhibition of SrC kinase activity and induction of tumor cell apoptosis.In compared the PCNA and SrC expression between control mice and ZD6474 treated mice,ZD6474-induced significant apoptosis of tumor cell,but a decrease in positive PCNA in the ZD6474 treat group compared with control.ZD6474 inhibit SrC Kinase expression in xenografied tumors,which is consisted with in vitro effects.SrC Kinase and its upregalation of Bcl-2-expression are essential mechanisms in imatinib-resistant K562 cell-lines.
