Perivascnlar adipose tissue (PVAT) has been thought to play an important role in regulating arterial tone in recent years.Perilipins associating with the lipid droplets in adipocytes suppress lipolysis in basal condition.Perilipin null (peri-/-) mice exhibit the phenotypes of increased basal lipolysis in adipocytes and reduced adipose mass.We investigated the vascular dysfunction associating with the PVAT in peri-/-mice.Vasoconstriction to phenylephrine decreased in the presence of PVAT in wt mice but was unaffected by PVAT in peri-/-mice, suggesting impaired PVAT anticontractile function.PVAT ofperi-/-mice showed increased superoxide production, as well as inflammatory Mac-3-positive cell infiltration.Our data suggest that perilipin is associated with PVAT function, and inflammation and oxidative stress are the main mechanisms for PVAT dysfunction in peri/ mice.
