L-ficolin mediates the phagocytosis of HCV through binding to TLR4 on macrophage

  We investigated whether L-ficolin, as a lectin, can play a role in the phagocytosis of HCV by macrophages through binding to TLR4 on the macrophages.In our study, Huh7.5.1 cells were transfected with pCMV-E1 or pCMV-E2, cell-surface expression of HCV envelope glycoprotein E1 and E2 were confirmed by Flow cytometry, Real-time PCR and Western Blot.Through CD14 positive magnetic bead selection, we got monocytes, which were then stimulated with M-CSF and induced to marcophages, from PBMC of healthy honors.Then, we used Flow cytometry to investigate the function of L-ficolin in the phagocytosis of Huh7.5.1-E1 or Huh7.5.1-E2 by macrophage.After that, the role of L-ficolin in peritoneal macrophages of mice were detected by the same method.
