Isonucleosides are a novel class of nucleoside analogues,in which nucleosides are linked to the different position ofribose other than C-1 position.When modified oligonucleotides with isonucleosides,it was proved that the regional conformation around the incorporated sites were changed.Here,D-/L-Isonucleoside,as a pair of enantiomer,were incorporated separately into the siRNA,in order to investigate the connection between the regional conformation alteration of siRNA and the physicochemicaland biological potency.Through computer simulation and thermo stability test,it was found that L-isonucleoside incorporation affected the regional conformation significantly,while D-isonucleoside incorporation affected slightly.
