Identification of Aptamers through Cell-SELEX for Targeting Primary Colon Adenocarcinoma

  Colon adenocarcinoma is one of the most deadly human cancers due to its strong ability of invasion and metastasis.The development of diagnostic and therapeutic strategies against metastasis-related molecular targets is critical for reducing death rates.In this work,we have developed a new aptamer against primary colon cancer cells SW480 as positive cells,and secondary colon cancer cells SW620 from the same patient metastatic site lymph node were used as negative cells.After 15 rounds of selection by cell-SELEX(Systematic Evolution of Ligands by Exponential enrichment)technique,one of the selected aptamers,151,with a dissociation constant(Kd)in nanomolar range against target cells SW480 was obtained.It can also specifically recognize other tested primary colon cancer cells and does not recognize other tested secondary colon cancer cells.The binding target of the selected aptamer has been preliminarily determined as a membrane protein on the cell surface.The aptamer generated in this work by cell-SELEX can selectively recognize primary colon cancer cells and holds potential applications in metastatic colon adenocarcinoma mechanism research,diagnosis and treatment.
