The innate immune system is equipped with sensitive and efficient machineries to provide an immediate, first line defense against infections.Toll-like receptors (TLRs) are an integral part of the innate immune defense system and trigger an innate immune response by activating signaling pathways that are dependent on interleukin-1 (IL-1) receptor-associated kinase (IRAKs) ,a serine/threonine-specific kinase (IRAK1, AK2, IRAK-M and IRAK4).TLRs recognize microbial components and associate with adaptor proteins lead to the recruitment and activation of IRAks family, which positively or negatively regulate a complex signaling cascade leading to the activation of NFκB and mitogen-activated protein kinases (MAPK) and to the production of defense molecules such as inflammatory cytokines, including interleukin-6 (I16) ,tumor necrosis factor (Tnfa) and KC.whether IRAK2 is catalytically active is still unclear. So we further restored BMMs(Bone marrow derived macrophages) from IRAK2 deficient mice with wild type。kinase defective of IRAK2 construct and test its function and kinase activity in TLRsignaling pathway .
