Biodegradable nanospheres or microsphere(MS) has been widely used as a sustained release system.In this research,Injectable sustained-release pcDNA3-GRF(1-32) microspheres are prepared by double emulsion-in liquid evaporation process using biodegradable ploy lactic-co-glycolic acid (PLGA) as carrier.The entrapment efficiency and mean particle size thus prepared are 89.79% and 2.41μm,respectively.The microspheres are injected into mice tibialis muscle.After 30 days,accumulative increased body weight in the group injected with pcDNA3-GRF(1-32) microspheres is significantly higher (20.50%,20.60% and 26.70%) than those group injected with naked plasmid,mixture of microspheres and plasmid and saline; PCR and RT-PCR taking DNA and RNA from injected muscle as template showes that the expression level of GRF gene on the group injected with pcDNA3-GRF(1-32) microspheres is the highest.Rabbits are injected with pcDNA3-GRF-MS,naked pcDNA3-GRF,blank MS,saline in tibialis muscle.
