Jadomycin B, an atypical angucycline polyketide, is synthesized by Streptomyces venezuelae ISP5230 under stress conditions.JadH, a bifunctional post-PKS tailoring enzyme, plays an important role in the biosynthesis of jadomycins.JadH catalyzes the C12-hydroxylation and C4a,12b-dehydration of 2,3-dehydro-UWM6, while most of its homologues, such as PgaE,CabE and UrdE, catalyze two hydroxylation reactions at C12 and C12b.In order to identify the key amino acids for the catalyzing activity of JadH, potential key residues were selected as mutational targets through constructing and analyzing the modeling of JadH binding to its substrate, as well as multiple sequence alignment with its homologues.
