波卓链霉菌基因簇克隆与异源表达

波卓霉素是波卓链霉菌(Streptomyces bottropensis)产生的,由核糖体负责合成的肽类抗生素,具有抗革兰氏阳性菌及支原体的功能,尤其重要的是具有anti-MRSA(抗甲氧西林金黄色葡萄球菌)和anti-VRE(抗万古霉素肠球菌)活性.其作用机制与已知抗生素没有交叉抗性,是一个具有重要前景的候选抗生素药物.由于波卓链霉菌遗传操作比较困难,有必要建立一个高效的基因中断体系,用于研究波卓霉素生物合成途径.柯斯质粒pJTU2554是整合型质粒,由该质粒构建的基因组文库可以实现基因簇的异源表达.因此,可以利用PCR-targeting系统在大肠杆菌中实现对基因簇中各个基因的高效敲除,然后将敲除基因后的质粒转移到模式链霉菌中,进一步研究基因簇中各基因功能及其生物合成途径.
