内质网应激状态下Nrf2细胞防御系统的激活及其对斑马鱼肝脏发育的影响

  有害物质和细胞压力在人体内的聚积,直接危害人类的健康.生物体通常会通过细胞防御系统的激活来对抗,减少机体损伤.Nrf2-Keap1 系统是一个非常重要细胞防御保护系统,它能对多种环境因子做出应答,启动抗氧化蛋白元件,调控抗氧化蛋白和Ⅱ相解毒酶的表达,在细胞的防御保护中发挥重要作用.Nrf2 系统在脊椎类动物中高度保存,在进行Nrf2 活性表达异常的人工变异斑马鱼筛选实验中,我们发现一组变异系统(it768).该系统在无任何外来刺激状下,仍可将Nrf2系统激活.证明了内在性的刺激对Nrf2 系统的作用.进一步的研究证明,这种内在性的刺激为内质网应激反应.
