Objective Tyro 3 is one of receptor tyrosine kinases in the TAM (Tyro 3, AXL and Mer) family, which is preferentially expressed in the neocortex and CA1, CA3 of hippocampus, and mainly associated with neurons.However, the relationship between tyro 3 and β-Amyloid (Aβ) is unreported.This study is to investigate the relationship between Tyro 3 and Aβ production.Methods We transfected APP plasmid to HEK293 cells and then selected by G418 to get HEK293 cells stably expressing APP.CMV-Tyro 3-CFP and CMV-GFP reporter plasmids were used to transfect the cells mentioned above, respectively.Meanwhile, the cells were treated with Gas6, which is the native ligand of Tyro 3.The supernatant Aβ was detected by ELISA.Results We observed that the production of Aβ was decreased in HEK293 cells transfected with CMV-Tyro 3-CFP compared with that of CMV-GFP transfected cells.Interestingly, the effect was reversed by treatment of Gas6 to the cultures.Conclusion In summary, our studies demonstrate that overexpression of Tyro 3 has the potential to reduce the generation of Aβ.Moreover, the effect of Tyro 3 on the production of Aβ is independent of Gas6.
