Identification of a halophilic actinobacterium Salininema proteolyticum 438HGGQ3-1 from rhizosphere

  During a previous bioprospecting of pharmaceutical actinomycetes in saline-alkali soil in Korla,Xinjiang,north-west China,strain 438HGGQ3-1T was isolated from a rhizosphere soil sample collected around the medicinal plant Lycium ruthenicum.The iaolate shared the highest levels similarity of 16S rRNA gene sequence with Salininema proteolyticum Miq-4T (99.80 %),Paraglycomyces xinjiangensis TRM 49201T (99.66 %) and Salilacibacter albus (96.00 %).Lower similarities (<93.54 %) were found with all other type strains in genera affiliated with family Glycomycetaceae.The isolate contained meso-diaminopimelic acid as the diagnostic diamino acid and glucose and ribose as the whole-cell sugars.The phospholipids consisted of phosphatidylethanolamine,diphosphatidylglycerol,phosphatidylglycerol,phosphati-dylinositol mannoside,phosphatidylinositol and two phosphoglycolipids.The major menaquinones were MK-9(H4),MK-10(H2) and MK-10(H4).The major fatty acids are anteiso-C15∶0,anteiso-C17∶0 and iso-C16∶0.The levels of DNA-DNArelatedness with S.proteolyticum IBRC-M 10908T (99.80 %) and P.xinjiangensis CCTCC AA 2013002T were all above 70 %,the cut-off value considered to be the threshold for the definition of genomic species.Based on the evidence from polyphasic taxonomic study,the isolate was identified as a strain of the species Salininema proteolyticum.
