In the Human Protein Atlas project antibodies to human proteins are systematically generated to be used in the creation of a publically available database of protein expression and localization in human normal and cancer tissues.The workflow has been set up in a modular fashion with antigen design, cloning, protein production, affinity purification and validation of polyclonal antibodies.To date more than 23000 antibodies have been generated and practically all genes have been initiated in the pipe-line.In this quest we have noted a positive correlation between solubility of the recombinant immunogen and acquired amount of antibody.Furthermore has a massive epitope mapping been performed to make a thorough investigation of the sequence ofepitope regions and silent antigenic regions.An iteration scheme has been set-up to handle the genes which fail in any of the steps to generate a validated antibody in order to make complete gene coverage in the atlas of the human proteome feasible.
