  How the blood vasculature and organs integrate is an important issue in development as well as in disease.In teleosts, the developing head kidney, composed of the pronephric glomerulus and the interrenal gland, interacts closely with the dorsal aorta (DA) during organogenesis and angiogenesis.We found in zebrafish that the DA-head kidney interaction was disrupted in the endodermless sox32 mutant and morphant.In contrast, the head kidney in Sox32-deficient embryo remained tightly associated with posterior cardinal vein (PCV), while both tissues demonstrated severe convergence defects.Rescued endoderm formation in the sox32 morphant by a simultaneous knockdown of Odd-skipped-related 1 significantly alleviated convergence defects of head kidney, indicating the importance of endoderm for patterning migration of the head kidney.Surprisingly, the severely defective convergence of head kidney tissues in the sox32-deficient embryo was suppressed by either the cloche (clo) mutation or the knockdown of clo-regulated genes which led to a disruption of axial vasculature.It suggested that head kidney tissues converged toward the midline even in the complete absence of endoderm and endothelium, thus the severely defective convergence of head kidney in the endodermless embryo could be due to a spatial restriction by the venous endothelium.Our findings thus indicated a role of venous vasculature for patterning the head kidney.
