TRAIL receptor agonists have been demonstrated as effective tumor therapeutics.Monoclonal antibodies against TRAIL receptor 1 or 2, such as Mapatumumab, Lexatumumab from Human Genome Sciences, Conatumumab from Amgen and LBY 135 from Novartis, have been passed through pre-clinical scrutiny and undergone clinical trials.In this report, we have screened out one scfv against TRAIL receptor 1 and one against TRAIL receptor 2 among around forty candidates respectively.Preliminary experiments showed that they can induce apoptosis in Colo 205, A549 and HCT116 cell lines at a nanomolar scale.In order to improve their affinity and efficacy, we pentamerized them as combodies using a coiled-coil domain from human cartilage oligomeric matrix protein(Asp29-Gln76, COMP48).
