Recently, one new kind of dual-functional mixed-mode separation media was synthesized to be used for the protein separation not only with IEC mode, but also with HIC mode called 2D column.The 2D column exhibits excellent resolution, selectivity, and retention of proteins and functions in two retention mode including HIC and WCX, which can be used to be instead of two commercial availabe columns for the fast separation of proteins, such as WAX and HIC columns.In this study, the refolding of the reduced/denatured lysozyme was investigated initially by 2D-LC with one single 2D column in WCX and HIC modes, respectively.
