Proteomic Analysis of Heat Stress-Responsive Proteins during Rice Grain Filling

  Heat stress is an increasing harmful effect on rice growth and development, especially at flowering and pollinating stage.The flowering rice plants (Oryza sativa L.var Nipponbare) were subjected to a temperature of 42 ℃ for 1 h and inflorescences were harvested after heat stress treatment.To identify the heat stress-responsive proteins, total proteins were separated by two-dimensional electrophoresis (2-DE) and visualized by Coomassie Brilliant Blue staining.A total of 23 protein spots were significantly changed, and all of them were identified by mass spectrometry analysis.The identified proteins were categorized into classes related to heat shock proteins, energy and metabolism, redox homeostasis, disease resistance, and senescence.The function of some of the responsive proteins remains unknown.The function of these identified proteins will be investigated by isolating the T-DNA mutants, RNAi or over-expression transgenic plants, in which the encoding genes are mutated or over-expressed.
