Introduction:Randomly Amplified Polymorphic DNA Analysis (RAPD-PCR) is one PCR technique which was developed to detect the specific 770 bp DNA product of Salmonella spp.by using a single short oligonucleotide primer This method is very rapid and sensitive. It does not distinguish between living and dead cells because DNA of some treated cells by mild conditions can give positive results. This limitation leads to overestimate the number of living cells. The aim of this study was to evaluate the effect of various strong lethal conditions on the analysis of Salmonella Typhimurium and Salmonella Enteritidis by RAPD-PCR.
