目的:观察Cedemex对纳洛酮催促下的戒断大鼠不同脑区环磷腺苷(cAMP)和环磷鸟苷(cGMP)含量的影响。方法:设空白、正常Cedemex、停用吗啡、续用吗啡、Gedemex 8,4,2 g·kg~(-1)·d~(-1)、益安口服液组8个实验组,每组10只大鼠。以剂量递增法连续皮下注射吗啡7 d建立吗啡依赖大鼠模型,采用放射免疫方法测定中脑腹侧被盖区(VTA)、皮层和海马的cAMP和cGMP含量。结果:Cedemex对大鼠吗啡戒断症状具有明显的抑制作用(P<0.05);与空白组相比较,续用吗啡组能够使依赖大鼠的VTA、皮层和海马内cAMP含量显著增高,cGMP含量显著下降(P<0.05);短期使用Cedemex可使吗啡戒断大鼠VTA、皮层和海马内cAMP含量明显下降,cGMP明显增高(P<0.05)。结论:Cedemex可显著抑制大鼠吗啡戒断反应,其可能的机制与Cedemex能调节中枢cAMP和cGMP含量有关。
OBJECTIVE: To observe the effects of Cedemex on the content of cAMP and cGMP in different brain regions of anaesthetized rats induced by naloxone. METHODS: Eight experimental groups with blank, normal Cedemex, discontinued morphine, continuation of morphine, Gedemex 8, 4, 2 g·kg -1 ·d~(-1), Yian oral solution group, each group 10 rats. Morphine-dependent rats were established by continuous subcutaneous injection of morphine 7 days after dose escalation. The cAMP and cGMP contents in the ventral tegmental area (VTA), cortex and hippocampus were determined by radioimmunoassay. RESULTS: Cedemex had a significant inhibitory effect on morphine withdrawal symptoms in rats (P<0.05). Continuation of morphine treatment significantly increased the cAMP content in VTA, cortex, and hippocampus of rats that were dependent on rats compared with the blank group. Significantly decreased (P<0.05); short-term use of Cedemex significantly decreased cAMP content in VTA, cortex and hippocampus of morphine withdrawal rats, and cGMP increased significantly (P<0.05). Conclusion: Cedexex can significantly inhibit the morphine withdrawal in rats. Its possible mechanism is related to the ability of Cedemex to regulate the content of cAMP and cGMP in the central nervous system.
