乙酰胆碱酯酶(AChE)是调节生物神经功能的一种酶,多种化合物,如有机磷和多种重金属离子,是乙酰胆碱酯酶活性的抑制成分。利用实验室自己合成的有色酶底物,2—(2—acetoxy—5—methyl—phenylazo)—N—methyl—1,3—thiazolium methosulfate(AMPT),它的溶液是黄色。在AChE酶的作用下,AMPT水解,其溶液转变为兰色的HMPT溶液(2—(2—hydroxy—5—methyl—phenylazo)—N—methyll,3—thiszokuym methosalfate)。但是,当有酶的抑制物质存在时,AChE酶会被抑制而失去部分活性。因此,溶液的颜色因酶抑制物质的浓度从兰色向黄色变化。利用光纤颜色测定仪测定溶液颜色的变化,就测定了抑制物质的浓度。
Acetylcholinesterase (AChE) is an enzyme that regulates biological nerve function. A variety of compounds, such as organophosphorus and various heavy metal ions, are inhibitors of acetylcholinesterase activity. Its solution is yellow using 2- (2-acetoxy-5-methyl-phenylazo) -N-methyl-1,3-thiazolium methosulfate (AMPT), a chromogenic substrate synthesized by the laboratory itself. Under the action of AChE enzyme, AMPT is hydrolyzed and the solution is converted to blue-colored HMPT solution (2- (2-hydroxy-5-methyl-phenylazo) -N-methyll, 3-thiszokuym methosalfate). However, AChE enzymes are inhibited and partially lost in the presence of enzyme inhibitors. Therefore, the color of the solution varies from blue to yellow due to the concentration of the enzyme inhibitor. Using a fiber optic colorimeter to determine the color change of the solution, the concentration of the inhibitory substance was determined.
