乙烯诱导凤梨开花受植株年龄的影响。本研究根据从抑制差减杂交(SSH)文库中发现的c DNA片段,通过RACE技术得到EIN3(ethylene insensitive3)基因的全长c DNA序列。生物信息学分析表明,该基因与香蕉、水稻、玉米等多种植物中的EIN3同源,定名为Af EIN3。Af EIN3在凤梨植株各个部位的表达量不同,以叶片基部的表达量最高,花器官中最少;Af EIN3在不同年龄植株中的表达量不同,以成熟植株中表达量最高;乙烯处理后,在成株中,Af EIN3的表达量在1 h即达到峰值,然后逐渐下降;在幼株中,Af EIN3对乙烯的反应较迟缓,24 h时略有上升,2 d时达到一个峰值,然后下降,5 d时表达量最高。研究结果表明Af EIN3具有组织表达特异性,表达量与发育阶段有关,并受乙烯信号诱导,可能在乙烯诱导凤梨开花中起重要作用。
Ethylene induced pineapple flowering influenced by plant age. In this study, the full-length c DNA sequence of the EIN3 (ethylene insensitive3) gene was obtained by RACE based on the c DNA fragments found from the SSH library. Bioinformatics analysis showed that the gene was homologous to EIN3 in banana, rice, maize and other plants, and named Af EIN3. The expression level of Af EIN3 in different parts of pineapple was different, the expression level of the leaf was the highest, and the floral organs were the lowest. The expression level of Af EIN3 in different age plants was the highest, In adult plants, the expression of Af EIN3 peaked at 1 h and then decreased gradually. In young plants, the response of Af EIN3 to ethylene was slow, increased slightly at 24 h, reached a peak at 2 d and then decreased, The expression was highest at 5 d. The results showed that Af EIN3 has tissue-specific expression. The expression level of Af EIN3 is related to the development stage and induced by ethylene signal, which may play an important role in ethylene-induced flowering of pineapple.
