目的:优化地榆总皂苷的碱水解工艺,提高水解产物地榆皂苷化合物Ⅳ的含量。方法:采用正交试验设计,考察水解时间、氢氧化钠体积分数、料液比、水解温度对地榆总皂苷水解过程的影响。结果:影响水解的主次因素依次是料液比>氢氧化钠体积分数>水解温度>水解时间,优选的最佳工艺为料液比1∶80;氢氧化钠体积分数20%,水解时间3.5 h,水解温度82℃。结论:采用优化后的水解条件能够得到含量较高的地榆皂苷化合物Ⅳ,适合工业化生产。
Objective: To optimize the alkaline hydrolysis process of Ulmus pumila and to increase the content of hydrolysates Saponins Ⅳ. Methods: The effects of hydrolysis time, sodium hydroxide volume fraction, solid-liquid ratio and hydrolysis temperature on the total saponin hydrolysis of Ulmus pumila were studied by orthogonal experimental design. Results: The primary and secondary factors affecting the hydrolysis were the ratio of solid to liquid> sodium hydroxide volume fraction> hydrolysis temperature> hydrolysis time. The optimal process was as follows: the ratio of solid to liquid was 1:80; the volume fraction of sodium hydroxide was 20%; the hydrolysis time was 3.5 h, hydrolysis temperature 82 ℃. Conclusion: Saponins Ⅳ with higher content can be obtained by the optimized hydrolysis conditions, which is suitable for industrial production.
