目的:观察淫羊藿两种主要有效成分:淫羊藿苷(ICA)和淫羊藿多糖(EPS)对气管切开插管大鼠肺部炎症的不同作用。方法:将SPF级健康雄性SD大鼠随机分成正常组(A组)、气管切开插管模型组(B组)、淫羊藿苷用药组(C组)、淫羊藿多糖用药组(D组),制作气管切开插管模型,造模成功6h后,C组开始给予淫羊藿苷灌胃,D组给予淫羊藿多糖灌胃,A、B两组以等量生理盐水,每日一次。分别在1d、3d、7d三个时间段每组分别处死6只大鼠,观察各组肺组织病理学变化,并用Simith评分测定肺损伤情况;采用酶联免疫吸附法(ELISA)测定支气管肺泡灌洗液中TNF-α和IL-6的浓度。结果:(1)光镜下B、C、D三组大鼠肺组织病理三个时段均有不同程度的炎症等改变,Smith评分均高于A组;但C、D两组病变在三个时段均明显小于B组。Smith评分显示C组病变明显小于D组。(2)支气管肺泡灌洗液中B、C、D三组TNF-α、IL-6含量变化趋势基本一致,在不同时间段均明显高于A组,24h时达到峰值,随后下降。两者在三组含量比较,B组最高,其次D组。结论:ICA和EPS均能起到抗炎作用,而ICA明显优于EPS。
OBJECTIVE: To observe the different effects of icariin (ICA) and epimedium polysaccharide (EPS) on lung inflammation in tracheotomy rats after intubation. Methods: SPF healthy male Sprague-Dawley rats were randomly divided into normal group (group A), tracheotomy model group (group B), icariin group (group C), Epimedium polysaccharide group (group D) Group), making tracheotomy intubation model, model 6h success, C group began to give icariin gavage, D group given epimedium polysaccharide gavage, A, B two groups with the same amount of saline, each Day time. Six rats in each group were sacrificed at 1d, 3d and 7d respectively. The pathological changes of the lungs in each group were observed, and the lung injury was measured by Simith score. The levels of bronchoalveolar lavage fluid (BALF) were measured by enzyme-linked immunosorbent assay (ELISA) Concentrations of TNF-α and IL-6 in the washings. Results: (1) Under the light microscope, the lung histopathology of rats in groups B, C and D all had different degrees of inflammatory changes and the scores of Smith were higher than those in group A. The lesions in groups C and D were in three The time was significantly less than the B group. Smith score showed that the group C lesion was significantly less than the D group. (2) The changes of TNF-α and IL-6 in B, C and D groups in bronchoalveolar lavage fluid were basically the same, and were significantly higher than those in group A at different time points, reaching the peak at 24h and then decreasing. The three groups in the content comparison, B group highest, followed by D group. Conclusion: Both ICA and EPS play an anti-inflammatory role, whereas ICA is significantly better than EPS.
