ABA(Abscisic Acid,脱落酸)是一种抑制型植物激素。它的结构是3-甲基-5(1’-羟基-4’-氧-2’6’6’-三甲基-2’-环己烯)顺反型2,4-戊二烯酸,分子量264.32,含量为5—200ng·g~(-1)植物鲜重。1984年10月,我们研制成功 ABA 的 RIA 法,国内迄今未见类似报道。ABA 免疫原系用碳化二亚胺法,由 ABA 与 BSA(牛血清白蛋白)缩合产生,每个 BSA 分子上偶联4个ABA 分子。免疫原经 Freund 完全佐剂乳化,于兔背皮内多点(80—100穴)免疫,制成抗体。用~3H-ABA 竞争性放射分析试样的 ABA,与气液色谱一电子捕获器法相比,本法的灵敏度更高(可测 1ng·ml~(-1)至10pg·ml~(-1)),r=0.9997;专一性强;重复性好(100份试样,每份4次重复,变异系数仅为3.47%);标本回收率≥96.0%;可测定少至1~10mg 植物鲜重内的 ABA;分析快速简便,全过程可在6小时内完成。本法的各项指标均已达国际上同类工作的水平。目前,本法已被应用于植物营养、代谢与遗传生理等研究领域,收效甚佳。我们正在研制 ABA 放射免疫试剂盒(包括 ABA 抗体、标准 ABA、~3H-ABA 等试剂),准备向外提供。
ABA (Abscisic Acid) is an inhibitory plant hormone. The immunogen was emulsified with Freund’s complete adjuvant, and then immunized with multiple spots (80-100 points) in the dorsal skin of rabbits to make antibodies. The ABA of the samples was analyzed by ~ 3H-ABA competitive radioactivity. Compared with gas-liquid chromatography-electron capture method, this method has higher sensitivity (1ng · ml -1 to 10pg · ml -1 )), r = 0.9997; specificity; good repeatability (100 samples, each repeated 4 times, the coefficient of variation was only 3.47%); sample recovery ≥ 96.0%; measurable as little as 1 ~ 10mg plants ABA within the fresh weight; analysis quick and easy, the whole process can be completed within 6 hours. The indicators of this Law have reached the level of similar work in the world. At present, this law has been applied to the field of plant nutrition, metabolism and genetic physiology and other fields, with good results. We are developing ABA radioimmunoassay kit (including ABA antibody, standard ABA, ~ 3H-ABA and other reagents), ready to provide.
