目的比较利多卡因和维拉帕米对乳腺癌细胞抗肿瘤药物的增敏作用。方法应用微量细胞培养四氮唑(MTT)法检测乳腺癌细胞系MCF-7对三种抗癌药物5-FU、MMC和CDDP敏感性。观察利多卡因和维拉帕米对三种化疗药物的增敏作用,并比较两药增敏作用之差异。结果 MCF-7对三种化疗药物均敏感,并随药物浓度的升高存活率降低。利多卡因和维拉帕米与三种化疗药物合用后分别降低三种化疗药物的半数抑制浓度(IC50),利多卡因比维拉帕米增敏作用强(P<0.01),加入利多卡因的增效倍数分别为27.4、30.1、59.1,而加入维拉帕米的增效倍数分别6.09、11.2、5.47。结论 MCF-7对三种化疗药物(5-FU、MMC及CDDP)均敏感,利多卡因和维拉帕米对三种化疗药物均有增敏作用,利多卡因比维拉帕米的增敏作用强。
Objective To compare the sensitization effect of lidocaine and verapamil to antitumor drugs in breast cancer cells. Methods The sensitivity of breast cancer cell line MCF-7 to 5-FU, MMC and CDDP was tested by MTT assay. To observe the sensitizing effects of lidocaine and verapamil on the three chemotherapeutic drugs, and to compare the difference of sensitization between the two drugs. Results MCF-7 was sensitive to all three chemotherapeutics, and the survival rate decreased with the increase of drug concentration. The combination of lidocaine and verapamil with three chemotherapeutic agents decreased the IC50 values of the three chemotherapeutic agents, respectively. The lidocaine had a stronger sensitizing effect than verapamil (P <0.01) Because of the multiples of multiples were 27.4,30.1,59.1, while the added multiples of verapamil efficiency were 6.09,11.2,5.47. Conclusion MCF-7 is sensitive to three chemotherapeutic drugs (5-FU, MMC and CDDP), and lidocaine and verapamil sensitize all three chemotherapeutic drugs.
