目的探讨氟尿嘧啶类药物代谢酶-胸腺嘧啶核苷酸磷酸化酶(TP)和二氢嘧啶脱氢酶(DPD)在正常组织与肿瘤组织中表达的差异性,探究其与结直肠癌临床病理特征之间的关系。方法半定量逆转录聚合酶链反应(RT-PCR),对40例结直肠癌肿瘤组织和正常结直肠黏膜组织中TP和DPD mRNA进行检测。结果结直肠癌肿瘤组织TP mRNA表达(0.82±0.06)与正常组织(0.79±0.06)相比差异无统计学意义,P=0.065;结直肠癌肿瘤组织DPD表达(0.65±0.09),明显高于正常组织(0.41±0.07),P<0.01;肿瘤患者中<65岁患者TP酶表达较高;DPD表达高的肿瘤浸润程度更高。结论人结直肠癌组织中DPD表达明显高于正常组织;肿瘤组织中TP和DPD表达与结直肠癌恶性程度存在一定相关性。
Objective To investigate the difference of the expression of fluorouracil-metabolizing enzymes - thymidine phosphorylase (TP) and dihydropyrimidine dehydrogenase (DPD) between normal tissues and tumor tissues, and to explore its relationship with the clinicopathological features of colorectal cancer The relationship between. Methods The expressions of TP and DPD mRNA in 40 colorectal cancer tissues and normal colorectal mucosa tissues were detected by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR). The expression of DPD in colorectal cancer tissues was significantly higher than that of colorectal cancer tissues (0.65 ± 0.09) Normal tissue (0.41 ± 0.07), P <0.01; Tumor patients with <65 years old patients with higher expression of TP enzyme; DPD high expression of a higher degree of tumor infiltration. Conclusion The expression of DPD in human colorectal cancer tissues is obviously higher than that in normal tissues. There is a certain correlation between the expression of TP and DPD in tumor tissues and the malignant degree of colorectal cancer.
