分离马尾杉内生真菌,并筛选其具抑菌活性菌株。用组织块分离法分离纯化菌株,通过形态学观察对其进行初步鉴定,以大肠杆菌、枯草芽孢杆菌和金黄色葡萄球菌为指示菌,采用对峙法进行抑菌活性筛选。实验分离获得马尾杉内生真菌28株,其中叶4株,叶鞘12株,茎9株,根3株。革兰氏染色结果表明,28株菌中,17株菌的菌丝染色呈紫色(60.71%),11株菌染色呈红色(39.29%),各菌的菌丝形态和孢子形态各异。抑菌实验结果表明所得28株马尾杉内生真菌,有11株至少能对1种指示菌有抑菌作用,占总分离菌株的39.29%,其中3株对金黄色葡萄球菌有较强的抑菌活性。马尾杉中存在大量的内生真菌,有些内生菌能产生抑菌活性代谢产物,可以进一步对这些代谢产物开展研究,为微生物药物资源的开发提供前期的物质基础。
Endophytic fungi were isolated from Marigold forest and their antibacterial activity was screened. Strains were isolated and purified by tissue block method. The morphological observation was used to identify the strains. Escherichia coli, Bacillus subtilis and Staphylococcus aureus were used as indicator bacteria to screen the antibacterial activity. 28 strains of endophytic fungus were obtained from the experiment, including 4 leaves, 12 leaves, 9 stems and 3 roots. Gram stain results showed that among the 28 strains, 17 strains of mycelia were stained purple (60.71%), 11 strains stained red (39.29%), and the mycelium morphology and spore morphology of each strain were different. Bacteriostatic test results showed that 28 of the resulting endophytic fungi Mosaic fir, 11 strains of bacteria can inhibit at least one indicator bacteria, accounting for 39.29% of the total isolates, of which 3 strains of Staphylococcus aureus has a strong antibacterial active. There are a large number of endophytic fungi in the plant, some endophytes can produce antibacterial activity of metabolites, these metabolites can be further carried out research for the development of microbial drug resources to provide early material basis.
