目的建立黄芩总黄酮部位3种成分黄芩苷、汉黄芩苷、黄芩素的HPLC含量测定方法。方法采用HPLC法,Waters-C_(18)色谱柱(250 mm×4.6 mm,5μm),以甲醇-水-磷酸(47:53: 0.2)为流动相,等度洗脱,流速0.6 ml/min,检测波长280 nm,柱温为室温(24℃)。结果黄芩苷、汉黄芩苷、黄芩素平均回收率分别为99.70%、100.06%和100.85%,RSD分别为2.47%、3.27%、2.66%。结论3批样品测定结果表明,该方法简便、准确,可用于黄芩总黄酮部位3种成分的含量测定。
Objective To establish a HPLC method for the determination of baicalin, wogonin and baicalein, three components of total flavonoids from Astragalus membranaceus. Methods HPLC method was used on a Waters-C_(18) column (250 mm×4.6 mm, 5 μm) with a mobile phase of methanol-water-phosphoric acid (47:53:0.2). Equivalent elution was performed at a flow rate of 0.6 ml/min. The detection wavelength was 280 nm, and the column temperature was room temperature (24°C). Results The average recoveries of baicalin, wogonin, baicalein were 99.70%, 100.06%, and 100.85%, respectively, and the RSDs were 2.47%, 3.27%, and 2.66%, respectively. Conclusion The determination results of three batches of samples showed that the method is simple and accurate, and it can be used to determine the content of three components of total flavonoids in Astragalus membranaceus.
