目的 建立裸鼠胶质瘤动物模型。方法 采用瘤细胞悬液接种法和瘤组织块接种法制作裸小鼠皮下移植瘤 ,并研究肿瘤的病理组织学特征。结果 两种方法所建肿瘤模型其成功率均为 10 0 % ,肿瘤生长状况良好 ,裸鼠无明显恶异质变化 ,肿瘤病理形态学检查符合胶质瘤细胞的形态学特征 ,免疫组化证实体外培养的胶质瘤细胞和体内胶质瘤组织胶质纤维酸性蛋白均高表达。结论 该方法所建裸鼠皮下移植瘤模型能作为研究胶质瘤发病机制、生物学特性以及基因治疗的可靠动物模型
Objective To establish a nude mouse glioma animal model. Methods Tumor cell suspension inoculation method and tumor tissue inoculation method were used to make subcutaneous xenografts in nude mice and to study the histopathological features of the tumors. Results The success rates of the two tumor models were 100%. The tumor growth was good. There was no obvious malignant change in the nude mice. The pathological morphological examination of tumor matched with the morphological features of glioma cells. Immunohistochemistry confirmed Glioma cells cultured in vitro and in vivo glioma tissue glial fibrillary acidic protein were highly expressed. Conclusion The model of nude mouse xenografts established by this method can be used as a reliable animal model for studying the pathogenesis, biological characteristics and gene therapy of glioma
