目的:对河北汉族群体6个STR短串联重复序列基国频率研究,得到河北汉族群体D7S820、D19S253,D12S391,D5S818,D16S539,D8S1179基因座的群体遣传学数据。方法:随机抽取173例河北省汉族人群无血缘关系个体的静脉血。肝素抗凝.碘化钾提取DNA,应用PCR技术扩增上述6个短串联重复序列基因座,采用高分辨的聚丙稀酰胺凝胶垂直板电泳,银染显色分析。结果:6个基因虚在河北汉族人群中都具有遗传多态性。D7S820,D19S253,D12S391,D5S818,D16S539,D8S1179基因座的杂合度分别为:0.828,0.757,0.769,0.837,0.785,0.852。个体识别率分别为:0.914,0.919,0.94,0.909,0.917,0.944。非父排除率分别为:0.618.0.74,0.801.0.557,0.655,0.696。遗传多态性信息量分别为:0.771,0.76,0.762,0.708,0.776,0.794。结论:上述6个STR基因座基因频率分布与Hardy-Weinberg平衡吻合良好,在河北汉族群体中具有较高的非父排除率与个体识别率,在群体遗传学研究及法医学?
OBJECTIVE: To study the frequency of 6 STR short tandem repeats in Hebei Han population and to obtain the population deportment data of Hebei Han population D7S820, D19S253, D12S391, D5S818, D16S539 and D8S1179 loci. Methods: A total of 173 venous blood from unrelated individual in Han nationality of Hebei Province were randomly selected. Heparin anticoagulation. The DNA was extracted by potassium iodide. The six short tandem repeat loci were amplified by PCR and analyzed by high-resolution polyacrylamide gel vertical plate electrophoresis and silver staining. Results: Six genetic polymorphisms were found in Hebei Han population. The heterozygosity of D7S820, D19S253, D12S391, D5S818, D16S539 and D8S1179 loci were 0.828,0.757,0.769,0.837,0.785,0.852, respectively. The individual recognition rates were: 0.914,0.919,0.94,0.909,0.917,0.944. Non-father exclusion rates were: 0.618.0.74,0.801.0.557,0.655,0.696. The amount of genetic polymorphism information were: 0.771,0.76,0.762,0.708,0.776,0.794. Conclusion: The frequency distribution of the above 6 STR loci is in good agreement with Hardy-Weinberg equilibrium.
