目的:研究尖吻蝮蛇毒抑瘤组分I(AAVC-I)对人肺癌A549细胞增殖抑制及凋亡的作用。方法:采用MTT法检测不同浓度的AAVC-I对A549细胞的24h与48h抑制率;应用HE染色、Hoechst33258荧光染色,从形态学观察细胞凋亡;采用免疫组化的方法,检测bax蛋白表达的变化。结果:MTT显示,AAVC-I能抑制A549细胞增殖,呈时间和剂量依赖性;AAVCI处理24h后,镜下可见细胞核固缩,核深染及凋亡小体;免疫组化显示,随药物浓度增加,平均光密度值呈递增趋势,表明bax蛋白表达相应上调。结论:尖吻蝮蛇毒抑瘤组分I对人肺癌A549细胞具有抑制作用,并诱导其凋亡,可能与bax表达上调有关。
Objective: To study the effect of AAVC-I on proliferation and apoptosis of human lung cancer A549 cells. METHODS: MTT assay was used to detect the inhibition rate of AAVC-I on A549 cells at 24h and 48h. HE staining and Hoechst 33258 fluorescence staining were used to observe the apoptosis of the cells. The expression of bax protein was detected by immunohistochemistry. Variety. RESULTS: MTT assay showed that AAVC-I could inhibit the proliferation of A549 cells in a dose- and time-dependent manner. After 24 hours of AAVCI treatment, the nucleus pyknosis, deep nuclear staining and apoptotic bodies were observed under microscope. Immunohistochemistry showed that the drug concentration With increasing, the average optical density value showed an increasing trend, indicating that the expression of bax protein was correspondingly upregulated. Conclusion: Anti-tumor component I of Agkistrodon acutus has inhibitory effect on human lung cancer A549 cells and induces apoptosis, which may be related to the up-regulation of bax expression.
