目的研究β-榄香烯对人骨髓瘤RPMI-8226细胞增殖、周期和凋亡的影响。方法用噻唑蓝(MTT)法检测β-榄香烯对骨髓瘤RPMI-8226细胞增殖的影响;Annexin V/PI双标流式术检测β-榄香烯对骨髓瘤RPMI-8226细胞周期、细胞凋亡的影响;Hoechst33342/PI双染荧光显微镜观察β-榄香烯处理后骨髓瘤RPMI-8226细胞形态学变化。结果β-榄香烯对RPMI-8226骨髓瘤细胞生长具有显著抑制作用,呈浓度和时间依赖性。β-榄香烯处理后,与对照组相比G0/G1细胞比例显著升高,而S、G2/M细胞比例降低。β-榄香烯作用48 h可诱导骨髓瘤细胞凋亡,凋亡率随药物浓度而递增。结论β-榄香烯可有效抑制人骨髓瘤细胞增殖;其抑制作用与细胞周期阻滞和细胞凋亡激活有关。
Objective To study the effect of β-elemene on the proliferation, cell cycle and apoptosis of human myeloma RPMI-8226 cells. Methods MTT method was used to detect the effect of β-elemene on the proliferation of RPMI-8226 myeloma cells. The apoptosis of RPMI-8226 cells was detected by Annexin V / PI double-flow cytometry. Apoptosis; Hoechst33342 / PI double staining fluorescence microscope after β-elemene treatment myeloma RPMI-8226 cells morphological changes. Results β-elemene significantly inhibited the growth of RPMI-8226 myeloma cells in a concentration- and time-dependent manner. After treatment with β-elemene, the proportion of G0 / G1 cells was significantly increased, while the proportion of S, G2 / M cells was decreased compared with the control group. β-elemene induced apoptosis of myeloma cells for 48 h, and the apoptosis rate increased with the concentration of the drug. Conclusion β-elemene can effectively inhibit the proliferation of human myeloma cells; its inhibitory effect is related to cell cycle arrest and activation of apoptosis.
