目的探讨4’-甲醚-黄岑素(4’-MS)对人绒毛膜癌细胞凋亡的影响。方法采用流式细胞术、实时定量PCR法及RT-PCR技术体外观察4’-MS对JAR细胞的影响。结果流式细胞术分析,加药组细胞凋亡率随4’-MS浓度的增加而逐渐升高,G2/M期细胞比例显著升高。20、40 mg/ml 4’-MS作用48 h后c-myc及h TERT mRNA的表达量明显低于对照组(P<0.01)。RT-PCR结果显示,20 mg/ml和40 mg/ml 4’-MS作用后,人绒毛膜癌JAR细胞中Survivin mRNA表达显著下降,而Caspase 9、Caspase 3及p38 MAPK mRNA表达显著上升(P<0.05)。结论绒毛膜癌JAR细胞在4-MS的作用下发生凋亡的机制与降低c-myc及h TERT mRNA的表达、上调Caspase 9表达、Caspase 3表达、激活p38 MAPK信号通路及抑制Survivin表达相关。
Objective To investigate the effect of 4’-methyl-4-methionine on human choriocarcinoma cell apoptosis. Methods The effects of 4’-MS on JAR cells were observed by flow cytometry, real-time quantitative PCR and RT-PCR. Results Flow cytometry analysis showed that the apoptotic rate in the dosing group increased gradually with the increase of 4’-MS concentration, and the proportion of cells in G2 / M phase increased significantly. The expression of c-myc and h TERT mRNA in 20,40 mg / ml 4’-MS for 48 h was significantly lower than that in the control group (P <0.01). The results of RT-PCR showed that the expressions of Survivin mRNA and the mRNAs of Caspase 9, Caspase 3 and p38 MAPK were significantly increased in human choriocarcinoma JAR cells after treatment with 20 mg / ml and 40 mg / ml 4’-MS (P <0.05). Conclusions The mechanism of apoptosis of JAR cells induced by 4-MS is related to the decrease of c-myc and h TERT mRNA expression, up-regulation of Caspase 9, expression of Caspase 3, activation of p38 MAPK signal pathway and inhibition of Survivin expression.
