目的:探讨FADS1 rs174537基因多态性与侵袭性牙周炎患者血清蛋白的相关性,为侵袭性牙周炎的诊断和治疗提供一定的理论基础。方法:选择353名侵袭性牙周炎患者(Ag P组)和125名健康对照者(HP组)进行FADS1 rs174537位点基因型和血清生化指标的检测,比较分析不同FADS1 rs174537基因型患者的血清总蛋白(total protein,TP)、球蛋白(globulin,GLB)、白蛋白(albumin,ALB)、白球比(albumin/globulin,A/G)。结果:(1)Ag P组FADS1 rs174537位点G等位基因检出率显著高于HP组(68.1%vs.61.2%,P=0.046,OR=1.35,95%可信区间为1.00~1.83);Ag P组FADS1 rs174537位点GG基因型的检出率显著高于HP组(45.5%vs.34.4%,P=0.029,OR=1.60,95%可信区间为1.05~2.44);(2)Ag P组GG基因型患者的TP、GLB水平显著低于GT+TT基因型患者[TP(77.08±7.88)g/L vs.(79.00±4.66)g/L,P=0.007;GLB(28.17±7.63)g/L vs.(29.88±3.49)g/L,P=0.007],A/G则显著高于GT+TT基因型患者(1.72±0.22 vs.1.67±0.22,P=0.040),而ALB水平差异没有统计学意义(P>0.05),HP组不同基因型者的TP、GLB、A/G及ALB差异均无统计学意义(P>0.05);(3)Ag P组GT+TT基因型患者的TP、GLB较相同基因型的健康受试者显著升高[TP(79.00±4.66)g/L vs.(75.20±4.53)g/L,P<0.01;GLB(29.88±3.49)g/L vs.(26.55±2.94)g/L,P<0.01],A/G更低水平(1.67±0.22 vs.1.88±0.30,P<0.01),差异具有统计学意义,而ALB水平差异无统计学意义(P>0.05);Ag P组GG基因型患者与HP组GG基因型者相比,TP、GLB、A/G及ALB差异均无统计学意义(P>0.05)。结论:FADS1 rs174537位点与侵袭性牙周炎的易感性密切相关,在侵袭性牙周炎患者中,该位点GG基因型与低TP、GLB、高A/G有关;FADS1 rs174537位点的GG基因型可能通过降低牙周防御机能,促进炎症反应,从而成为侵袭性牙周炎的重要危险因素之一。
Objective: To investigate the relationship between FADS1 rs174537 gene polymorphism and serum proteins in patients with aggressive periodontitis, and to provide a theoretical basis for the diagnosis and treatment of aggressive periodontitis. Methods: The genotypes and serum biochemical indexes of FADS1 rs174537 were detected in 353 patients with aggressive periodontitis (Ag P group) and 125 healthy controls (HP group). The serums of patients with different FADS1 rs174537 genotypes were compared Total protein (TP), globulin (GLB), albumin (ALB) and albumin / globulin (A / G). There was no significant difference in TP, GLB, A / G and ALB between different genotypes in HP group (P> 0.05). (3) GT + TT The levels of TP and GLB in genotype patients were significantly higher than those in healthy subjects with the same genotype [TP (79.00 ± 4.66) g / L vs. (75.20 ± 4.53) g / L, P <0.01; GLB (29.88 ± 3.49) (26.55 ± 2.94) g / L, P <0.01], A / G lower level (1.67 ± 0.22 vs.1.88 ± 0.30, P <0.01), the difference was statistically significant, while the ALB level difference (P> 0.05). Compared with GG genotype in GG genotype of Ag group and HP genotype, TP , GLB, A / G and ALB had no statistical significance (P> 0.05). CONCLUSIONS: FADS1 rs174537 locus is closely related to the susceptibility to aggressive periodontitis. In patients with aggressive periodontitis, the GG genotype at this locus is associated with low TP, GLB, high A / G, and FADS1 rs174537 GG genotype may be one of the important risk factors of invasive periodontitis by reducing periodontal defensive function and promoting inflammatory reaction.
