目的:研究神经生长因子(NGF)急性毒性及长期毒性。方法:小鼠急毒按最大耐受量测定法进行大鼠长期毒性试验时分低(5000BU/kg)、中(7070BU/kg)、高(10000BU/kg)3个试验组和1个生理盐水对照组;im,共90天。部分动物给药结束后继续观察14天。结果:小鼠一次ip、im、ivNGF的最大耐受量均>25000BU/kg,大鼠连续imNGF90天,给药期间、给药结束及停药14天时血液学、血液生化、病检均无异常所见,不影响大鼠行为和生长发育。结论:NGF对小鼠一次性iv、ip、im的最大耐受量大于25000BU/kg,90天肌肉内注射给药,对大鼠无明显毒性。
Objective: To study the acute toxicity and long-term toxicity of nerve growth factor (NGF). Methods: The acute toxicity of mice was determined according to the method of maximum tolerated dose. Long-term toxicity test was performed in rats at low dose (5000BU / kg), medium dose (7070BU / kg) and high dose (10000BU / Control group; im, a total of 90 days. Results: The maximum tolerated dose of ip, im and ivNGF in mice was> 25000BU / kg and the imNGF was continuously administered to rats for 90 days.
