More than 40 years ago, Yabuta discovered the gene exchange between genes in higher plants and fungi. Based on this principle, Andrea et al. began research on the endogenous micro-organisms of Taxus chinensis in 1992, focusing on the study of paclitaxel production in this tree, with a view to finding a new biological resource for paclitaxel and addressing the increasing demand for paclitaxel. Over the past two years, the researchers isolated more than 300 strains of fungi from healthy and diseased yew bark, needles and roots. First, the sample was placed in a pure aqueous agar medium. After the colony was formed, it was inoculated into the fungal medium. The first fermentation broth was supplemented with 2% yew needle leaf juice as an essential precursor resource reservoir or as a paclitaxel biosynthetic enhancer, and the subsequent fermentation broth was added only strictly limited combinations.
