应用凝胶过滤高效液相色谱,测定了两个两亲性多肽Amp1和Amp2进入磷脂酰甘油/磷脂酰胆碱脂双层的表观分配常数,并利用三硝基苯磺酸分析研究了与脂质体结合的多肽的氨基暴露状况。由结果推测,处于膜结合状态的多肽的氨基端是暴露于水相的;Amp1与脂双层相互作用强于Amp2,一方面表现为Amp1比Amp2埋膜较深,另一方面表现为Amp1与脂双层的结合能力比Amp2强,而主要表现在于后者。此外也发现两个多肽在缓冲液中处于几乎不存在暴露的氨基的聚合状态。
The apparent partitioning constants of two amphipathic peptides Amp1 and Amp2 into the phosphatidylglycerol / phosphatidylcholine lipid bilayer were determined by gel filtration HPLC, and the interaction between Amino-exposed conditions of liposome-conjugated polypeptides. The results suggest that the amino-terminus of the membrane-bound polypeptide is exposed to the aqueous phase. The interaction between Amp1 and the lipid bilayer is stronger than that of Amp2. On the one hand, the expression of Amp1 is deeper than that of Amp2; on the other hand, Lipid bilayer binding ability than Amp2, but mainly in the latter. In addition, two polypeptides were also found to be in a polymerized state where there is almost no exposed amino group in the buffer.
