目的通过小分子干扰RNA(siRNA)沉默livin的表达,探讨其对胶质瘤U251细胞增殖及凋亡的影响。方法制备livin siRNA重组腺病毒后感染U251细胞,采用RT-PCR、蛋白质印迹法检测livin基因的干扰效果,用MTT法和流式细胞术检测U251细胞的增殖和凋亡情况。同时观察重组腺病毒对荷瘤小鼠的治疗效应。结果重组腺病毒Ad-livinsiRNA能有效抑制U251细胞的livin mRNA和蛋白水平的表达;抑制U251细胞增殖,促进凋亡;在小鼠体内有效抑制U251细胞的生长和提高荷瘤小鼠的平均生存期(P<0.05)。结论用livin siRNA重组腺病毒介导抑制livin基因的表达,可抑制U251细胞增殖,促进凋亡,为胶质瘤的治疗提供了新策略。
Objective To silence the expression of livin by small interfering RNA (siRNA) and investigate its effect on the proliferation and apoptosis of glioma U251 cells. Methods U251 cells were infected with livin siRNA recombinant adenovirus. The interference effect of livin gene was detected by RT-PCR and Western blotting. The proliferation and apoptosis of U251 cells were detected by MTT and flow cytometry. At the same time observe the therapeutic effect of recombinant adenovirus on tumor-bearing mice. Results Recombinant adenovirus Ad-livinsiRNA could effectively inhibit the expression of livin mRNA and protein in U251 cells, inhibit the proliferation and promote the apoptosis of U251 cells, effectively inhibit the growth of U251 cells and increase the average survival of tumor-bearing mice (P <0.05). Conclusions Livin siRNA recombinant adenovirus can inhibit the expression of livin gene, inhibit the proliferation and promote the apoptosis of U251 cells, and provide a new strategy for the treatment of gliomas.
