目的:从受体水平上探讨儿童单纯肥胖症脂质代谢紊乱情况及其与微量元素之间的关系。方法:用放射配体分析法对31例单纯肥胖儿童及按性别年龄11配对的31例正常健康儿童进行外周血单个核细胞(PBMC)低密度脂蛋白受体(LDL-R)检测,同时测定血清锌(Zn)、铜(Cu)、铁(Fe)水平,并利用体外细胞培养观察Zn、Cu、Fe对PBMC上LDL-R表达的影响。结果:与正常对照组相比,肥胖组PBMC上LDL-R位点数明显减少(P<0.001),而血清Zn水平显著升高、Cu/Zn比值降低(P<0.05),且Cu与LDL-R呈正相关(P<0.01);体外实验发现加Cu培养时PBMC上LDL-R位点数较空白对照明显增加(P<0.01)。结论:儿童单纯肥胖症LDL-R存在表达障碍,机体微量元素稳态失调在其发生中可能起着一定的作用
OBJECTIVE: To investigate the disorder of lipid metabolism in children with simple obesity and its relationship with trace elements at receptor level. Methods: LDL-R was detected in peripheral blood mononuclear cells (PBMCs) in 31 children with obesity and 31 normal healthy children paired by sex and age 11 by radioligand analysis. (Zn), copper (Cu) and iron (Fe) were determined. The effects of Zn, Cu and Fe on the expression of LDL-R on PBMC were observed by in vitro cell culture. Results: Compared with the normal control group, the number of LDL-R loci in obese group was significantly decreased (P <0.001), while the level of serum Zn was significantly increased and the ratio of Cu / Zn was decreased (P <0.05) There was a positive correlation between Cu and LDL-R (P <0.01). In vitro, the number of LDL-R loci on PBMC was significantly increased compared with control (P <0.01). Conclusion: The expression of LDL-R in childhood obesity is impaired, and the homeostasis of trace elements may play a role in its occurrence
