本研究选用陕西境内5个采样点采集的30个菜粉蝶个体,用AFLP技术分析了5个种群的遗传多样性。凝胶图经染色和分析发现总位点数为540个,其中504个为多态性位点,36个共有位点,具多态性的条带百分率为93.33%。结果表明:AFLP技术适合应用于菜粉蝶遗传多样性分析;荧光标记法在AFLP分析中具有较高的检测效率;南北坡5个地点种群的遗传多样性分化显示出山脉对菜粉蝶的隔绝作用。
In this study, 30 individuals of Pieris rapae collected from 5 sampling sites in Shaanxi Province were selected and the genetic diversity of 5 populations was analyzed using AFLP technique. Gel staining and analysis showed that the total number of loci was 540, of which 504 were polymorphic loci and 36 shared loci. The percentage of bands with polymorphism was 93.33%. The results showed that the AFLP technique was suitable for the genetic diversity analysis of Pieris rapae. Fluorescent labeling showed high detection efficiency in AFLP analysis. The genetic diversity of five populations on the north and south slopes showed the isolation of the cabbage butterfly from the mountains.
