通过考察莪术醋制前后对由肝脏分泌的胆汁中内源性代谢产物的影响,以期探讨莪术醋制对胆汁内源性代谢物的影响。分别制备生、醋莪术醇提液及生理盐水溶液,灌胃给药0.5 h后做胆总管插管引流术收集12 h内大鼠胆汁;胆汁样品1∶3乙腈沉淀蛋白后进行UPLC-TOF-MS分析;采用单维统计与多元统计相结合,Peak View软件与网络数据库相比对的数据分析方法,鉴定潜在生物标记物。醋制前后莪术提取物对大鼠胆汁代谢谱具显著影响,鉴别出生、醋莪术给药组的大鼠胆汁中具有显著性差异的13个代谢物;再结合化合物网络数据库进行鉴定,得到8个代谢物;进一步把给药组与空白组进行t检验单维统计分析,得到与空白组比较具显著性差异的7个代谢物,推定此7个代谢物为生、醋莪术给药后大鼠胆汁的潜在生物标记物,其中6个潜在生物标记物在醋品组中显示上调,其与磷脂代谢、脂肪代谢、胆汁酸代谢及与调控N-酰基乙醇胺水解反应平衡作用相关的代谢调控相关,提示通过大鼠胆汁内源性代谢物的差异探讨莪术醋制增效的机制。
By studying Curcuma vinegar before and after the secretion of endogenous liver bile metabolites, in order to explore the Curcuma vinegar on bile endogenous metabolites. Respectively, preparation of raw, vinegar Curcuma alcohol extract and saline solution, 0.5 h after intragastric administration of common bile duct catheterization to collect rat bile within 12 h; bile samples 1: 3 acetonitrile precipitated protein UPLC-TOF- MS analysis; using single-dimensional statistical and multivariate statistics, Peak View software compared with the network database data analysis methods to identify potential biomarkers. Vinegar before and after extract of Curcuma significantly affects rat bile metabolism profile to identify the birth, Curcuma Rhizoma medication group of rats with significant differences in the bile of 13 metabolites; combined with the compound network database identified 8 Metabolites were further analyzed by t-test. The results showed that there were 7 metabolites with significant difference compared with blank group, and the 7 metabolites were assumed to be viable. Rats administered with Curcuma phaeocaulis The potential biomarkers of bile, of which six potential biomarkers showed upregulation in the vinegar group, were associated with metabolic regulation of phospholipid metabolism, lipid metabolism, bile acid metabolism and the balance of the regulation of N-acyl ethanolamine hydrolysis reaction, It is suggested that the mechanism of the synergism of Curcuma vinegar through the difference of endogenous metabolites of bile in rats.
