目的探讨蜕膜巨噬细胞凝血酶敏感蛋白-1(TSP-1)、白介素-10(IL-10)及干扰素-γ(IFN-γ)的表达。方法对2005年7月至2006年7月在上海交通大学附属仁济医院计划生育门诊要求人工流产的正常妊娠妇女20例(A组)和10例原因不明复发性流产(URSA)患者(B组)检测蜕膜TSP-1mRNA、蜕膜巨噬细胞表面TSP-1的表达及蜕膜巨噬细胞分泌IL-10和IFN-γ的量,并在培养液中加入相应刺激物,观察细胞因子分泌量的变化。结果与A组相比,B组蜕膜TSP-1mRNA表达明显降低(P<0·05)。蜕膜巨噬细胞TSP-1表达水平明显降低(P<0·01)。蜕膜巨噬细胞分泌IL-10的量显著降低(P<0·05);在培养液中加入各种干预因素对两组病例蜕膜巨噬细胞分泌IFN-γ无显著影响;加入TSP-1后,两组蜕膜巨噬细胞分泌IL-10的量均增加(P<0·05);加入抗TSP-1后A组蜕膜巨噬细胞分泌IL-10的量降低(P<0·05)。结论URSA患者蜕膜TSP-1表达下降,且蜕膜巨噬细胞分泌IL-10的量降低,可能是导致URSA发病的原因。TSP-1可能在调控蜕膜巨噬细胞分泌IL-10的过程中起重要作用。
Objective To investigate the expression of thrombospondin-1 (TSP-1), interleukin-10 (IL-10) and interferon-γ (IFN- γ) in decidual macrophages. Methods From July 2005 to July 2006, 20 pregnant women (group A) and 10 pregnant women with unexplained recurrent spontaneous abortion (URSA) in the family planning outpatient department of Renji Hospital Affiliated to Shanghai Jiao Tong University (group B ) Was used to detect the expression of TSP-1 mRNA and the expression of TSP-1 on the surface of decidual macrophages and the amount of IL-10 and IFN-γ secreted by decidual macrophages. Corresponding stimuli were added to the culture medium to observe the secretion of cytokines The amount of change. Results Compared with group A, the decidual TSP-1 mRNA expression in group B was significantly lower (P <0.05). Decidual macrophages TSP-1 expression was significantly lower (P <0.01). The amount of IL-10 secreted by decidual macrophages was significantly decreased (P <0.05). Adding various intervention factors to the culture fluid had no significant effect on the secretion of IFN-γ by decidual macrophages in both groups. Adding TSP- 1, the amount of IL-10 secreted by decidual macrophages in both groups increased (P <0.05); the amount of IL-10 secreted by decidual macrophages in group A decreased after addition of anti-TSP-1 (P <0 · 05). Conclusions The expression of TSP-1 in decidua of URSA patients is decreased and the amount of IL-10 secreted by decidual macrophages is decreased, which may be responsible for the pathogenesis of URSA. TSP-1 may play an important role in regulating the secretion of IL-10 by decidual macrophages.
