目的探讨多囊卵巢综合征(Polycystic ovarian syndrome,PCOS)患者子宫内膜“植入窗”期凋亡相关基因的差异性,并对部分基因进行了定位研究。方法将PCOS患者和对照组各6例共分为3组,分别取“植入窗”期的子宫内膜组织,应用基因芯片技术,检测其差异表达基因,对芯片数据进行处理和生物信息学分析,得到了差异表达的凋亡基因,并对下调的Fas相关死亡结构域蛋白(Fas-associated death domainprotein,FADD)基因运用反转录-PCR及免疫组织化学方法对进行了表达及定位研究。结果 PCOS 3组中均明显差异表达的基因有194条,有9条为凋亡相关基因,其中5个上调,4个下调;免疫组织化学研究发现,FADD免疫阳性物质位于PCOS患者“植入窗”期子宫内膜腺上皮细胞的细胞浆内。结论 PCOS患者“植入窗”期子宫内膜部分凋亡基因表达紊乱,是影响其植入率的原因之一。
Objective To investigate the differences of apoptosis-related genes in endometrium and implantation window of patients with polycystic ovarian syndrome (PCOS) and to locate some genes. Methods Six patients with PCOS and control group were divided into three groups. The endometrial tissues of the “implantation window” period were respectively harvested. Gene chips were used to detect the differentially expressed genes. According to the informatics analysis, differentially expressed apoptotic genes were obtained, and the expression and localization of down-regulated Fas-associated death domain protein (FADD) gene was detected by reverse transcription-PCR and immunohistochemistry the study. Results There were 194 differentially expressed genes in PCOS 3 group, 9 of them were apoptosis-related genes, of which 5 were up-regulated and 4 were down-regulated. Immunohistochemistry showed that FADD-immunopositive substances were located in PCOS patients Window “endometrial glandular epithelial cells within the cytoplasm. Conclusion The expression of some apoptotic genes in PCOS patients at the ”implantation window" stage is one of the reasons that affect the implantation rate.
